Abstract
Siklamen Myrsinaceae familyasına ait özellikle Akadeniz havzasın dağılmış 23 tür ile yayılış göstermektedir. Topalamda 23 türün 12’ si Türkiye’de doğal olarak yayılış göstermektedir ve 6 tanesi endemiktir. Türler arası melezleme yeni çeşitler elde etmek için önemli bir tekniktir. Siklamen türleri gibi türlerde melezleme sonrası abortif hybrid embriyo oluşumu gibi melezleme bariyerleri meydana gelmektedir. Melezleme sonrası oluşan abortif embriyoların rejenerasyonu için, yeni türlerin elde edilmesi amacıyla embriyo kurtarma tekniği uygulanmaktadır. Bu çalışmada ticari siklamen türü olan ‘Maxora’, Türkiye’de doğal olarak yayılış gösteren C. persicum ve C. coum türlerinde karşılıklı melezleme yapılmış ve embriyo kurtarma amacıyla tozlamadan 35. gün sonra ovül kültürü yapılmıştır. Çiçek tomurcuğu içerisinden ovüller izole edilmiş ve ½ MS + 2.0 mg/l 2,4-D + 0.8 mg/l 2iP ve ½ MS + 0.4 mg/l BA + 0.4 mg/l GA3 içeren besi yerinde kültüre alınmıştır. Eksplantlardan ½ MS 2.0 mg/l 2,4-D + 0.8 mg/l 2iP besi yerinde %30 oranında kallus oluşumu sağlanmıştır. Elde edilen kalluslardan embriyoya dönüşüm oranı % 19, bitkicik oluşumu ise %12 olarak belirlenmiştir İn vitro rejenerasyon çalışmaları sonucunda elde edilen bitkilerde aklimatizasyon oranı % 29.41 olmuştur.
