Keçilerde Klasik ve Uzun Etkili (LA) Sefkuinom Formülasyonlarının Farmakokinetiklerinin Karşılaştırılması

Abstract

Bu araştırmanın amacı sefkuinomun %2.5 (klasik) ve %7.5’luk (uzun etkili-LA) formülasyonlarının keçilerde kas içi (IM) uygulanmasını takiben farmakokinetiklerini karşılaştırmaktır. Araştırmada, 6 adet sağlıklı erkek Kilis keçisi kullanıldı. Çalışma, uygulamalar arası 15 günlük ilaç arınma süresini takiben iki aşamada çapraz farmakokinetik dizayna göre gerçekleştirildi. Çalışmanın birinci aşamasında 3 keçiye %2.5’luk diğer 3 keçiye de %7.5’luk sefkuinom formülasyonu IM yolla 2 mg/kg tek doz uygulandı. İkinci aşamada ise keçiler farklı formülasyonları alacak şekilde değiştirildi. Kan örnekleri uygulamayı takiben 0 (kontrol), 5, 10, 15, 20, 30 ve 45. dakikalar ile 1, 2, 3, 4, 5, 6, 8, 10, 12, 24, 48 ve 72. saatlerde alındı. Plazma sefkuinom konsantrasyonları yüksek performanslı sıvı kromatografisi (HPLC)-UV kullanılarak ölçüldü ve farmakokinetik parametreler non-kompartmantel analiz ile hesaplandı. Keçilere %2.5’luk sefkuinom uygulamasını takiben ortalama eliminasyon yarılanma ömrü (t1/2ʎz), eğri altında kalan alan (EAA0-son), plazma doruk konsantrasyon (Cdoruk) ve doruk konsantrasyona ulaşma süresi (Tdoruk) sırasıyla 9.230.45 saat, 15.171.23 saat*µg/mL, 2.630.15 µg/mL ve 1 saat idi. Sefkuinomun %7.5’luk formülasyonunun uygulaması sonrası t1/2ʎz’nin (62.355.06) uzadığı, EAA0−son (33.312.00) ve Tdoruk (6) değerlerinin arttığı ve Cdoruk’un (0.82  0.04) azaldığı belirlendi. Sonuç olarak %2.5’luk formülasyon ile %7.5’luk formülasyon karşılaştırıldığında sefkuinomun farmakokinetiğinde önemli farklılıklar olduğu belirlendi. Fakat keçilerde bakteriyel hastalık durumunda sefkuinomun klasik (%2.5) ve LA (%7.5) formülasyonlarının farmakokinetik ve farmakodinamik etkilerinin ortaya konulmasına ihtiyaç vardır.
