LAPAROTOMİ UYGULANAN SIÇANLARDA KALKON-İMİD KULLANIMININ OKSİDATİF STRES ÜZERİNE ETKİSİNİN DEĞERLENDİRİLMESİ

Abstract
Bu çalışmada, yeni sentezlenmiş olan Kalkon-imid türevinin laparotomi uygulanan sıçanlarda oksidatif stres üzerine etkisinin değerlendirilmesi amaçlanmıştır. Çalışmada 48 adet sağlıklı erkek wistar albino sıçan kullanılmıştır. Bu sıçanlardan kontrol grubuna herhangi bir deneysel işlem uygulanmamıştır. 1. ve 2. Şam grubuna %1 oranında DMSO intraperitoneal yolla verilip sıçanlar sırasıyla 7. ve 14. günde sakrifiye edilmiştir. Diğer gruplarda batın duvarı laparotomi ile açılmış ve 25-50 mg/kg-Kalkon uygulanmış ve 7. ve 14. günün sonunda sakrifiye edilmiştir. Sıçanların serumlarında Total Antioksidan Seviyenin (TAS) ve Total Oksidan Seviyesinin (TOS) ölçümü yapılmış ve olsidatif stres indeksi (OSİ) hesaplanmıştır. Grupların serum TAS, TOS ve OSİ düzeyleri karşılaştırılmış ve serum TAS, TOS ve OSİ değerleri arasında anlamlı fark bulunamamıştır. Bu bulgular bize, yara iyileşmesinin erken evrelerinde, bir flavanoid türevi olan kolkon-imid lokal kullanımının sistemik oksidatif stres üzerine bir etkisi olmadığını düşündürmektedir.

Abstract
The aim of this study was to evaluate the effect of a newly synthesised chalcone-imide derivative on oxidative stress in laparotomised rats. Forty-eight healthy male wistar albino rats were used in the study. No experimental procedure was applied to the control group of these rats. In the 1st and 2nd Damascus groups, 1% DMSO was administered intraperitoneally and the rats were sacrificed on the 7th and 14th day, respectively. In the other groups, the abdominal wall was opened by laparotomy and 25-50 mg/kg Chalcone was administered and sacrificed at the end of the 7th and 14th days. Total Antioxidant Status (TAS) and Total Oxidant Status (TOS) were measured in the serum of rats and oxidative stress index (OSI) was calculated. Serum TAS, TOS and OSI levels of the groups were compared and no significant difference was found between serum TAS, TOS and OSI values. These findings suggest that local use of colcon-imide, a flavanoid derivative, has no effect on systemic oxidative stress in the early stages of wound healing.
