Hindistan Cevizi Suyunun Serapias vomeracea’nin In vitro Gelişimine Etkileri

Abstract

Bu çalışma, Hindistan cevizi suyunun (HCS) Serapias vomeracea'de in vitro asimbiyotik tohum çimlenmesi, protokorm oluşumu ve organ gelişimi üzerine konsantrasyona bağlı (25, 50 ve 100 ml L-1) etkilerini karşılaştırmayı amaçlamaktadır. Farklı HCS konsantrasyonlarının tohum çimlenmesi ve protokorm oluşumu üzerindeki etkileri kültür ortamında sükroz varlığında ve yokluğunda test edilmiştir. Kültür ortamında HCS ve sükrozun eşzamanlı bulunması tohumların çimlenme oranını düşürdüğü için organogenez üzerine HCS'nin etkileri sadece sükroz yokluğunda denenmiştir. Kontrol ortamı ve sadece HCS içeren ortam en yüksek tohum çimlenme oranlarını vermesine rağmen sonuçlar arasında istatistiki fark bulunmamıştır. Bununla birlikte, tüm HCS konsantrasyonları kültür ortamında sükrozdan bağımsız olarak protokorm oluşumunu önemli ölçüde artırmıştır. En yüksek ortalama sürgün uzunluğu 100 ml L-1 konsantrasyonunda HCS uygulaması sonucunda kaydedilmiştir. HCS uygulamalarından sonra rizogenik cevap anlamlı derecede azalmış ve tüm HCS uygulamalarının kök uzamasını azalttığı bulunmuştur. Kök oluşumu artan HCS konsantrasyonları ile artmış ancak yine de kontrol değerinin altında kalmıştır. Kültürlerde yumru oluşumu gözlenmemiştir. Bu çalışma HCS'nin S. vomeracea tohumlarının in vitro çimlenmesinde sükroz yerine kullanılabileceğini önermektedir. Bununla birlikte, çalışmanın sonuçları HCS'nin S. vomeracea'de organ gelişimi üzerine sadece 100 ml L-1'den daha yüksek bir konsantrasyonda veya sükroz ile birlikte daha düşük konsantrasyonlarda kullanılması durumunda pozitif etkiler yaratabileceğini göstermiştir.
