Tricholoma fracticum Ekstresinin Hep3B Hücreleri Üzerindeki Sitotoksik Etkisi

Abstract

Kanser, insan sağlığı için en büyük tehditlerden birini oluşturmaktadır. İlaç direncinin kanser tedavisindeki temel sorunlardan biri olduğu düşünüldüğünde, doğal bileşiklerin kanser hücreleri üzerindeki etkilerine yönelik çalışmalar hızla artmaktadır. Yenilebilir mantarlar besin değerleri bakımından zengin olmaları nedeniyle fonksiyonel ve sağlıklı gıda kaynağı olarak değerlendirilmektedir. Yapılan çalışmalarda antikanser, anti-inflamatuvar, antioksidan ve antimikrobiyal etkiye sahip oldukları tespit edilmiştir. Ülkemizin birçok bölgesi ve Elazığ ilinde doğal olarak yetişen Tricholoma fracticum (Britzelm.) Kreisel besin olarak tüketilen yenilebilir bir mantardır. Bu çalışmada T. fracticum’un farklı konsantrasyonlardaki (1000µg/mL, 500 µg/mL, 250 µg/mL, 125 µg/mL ve 62.5 µg/mL) metanol ekstresinin 24 ve 48 saatte Hep3B hücrelerine karşı sitotoksik etkisinin belirlenmesi amaçlanmıştır. Sitotoksik etkinin tespit edilmesi için 3-(4,5dimetiltiazol-2-iyl)-2,5-difenil tetrazolyum bromür (MTT) yöntemi kullanılmıştır. Elde edilen sonuçlarda T. fracticum’un metanol ekstresi 1000 µg/mL konsantrasyonda 24. (%1.97±0.65) ve 48. saatte (%0.61±0.27) önemli sitotoksik etki göstermiştir. Mantarın metanol ektresinin Hep3B hücre hattına karşı 24. saatte IC50 değerinin ise 166.76±7.65 µg/mL ve 48. saatte 134.38±5.45 µg/mL olarak hesaplanmıştır. T. fracticum ekstresinin Hep3B hücre hattına karşı elde edilen sitotoksik sonuçlarının önemli olabileceğini ve Tricholoma türleri arasında daha az çalışılan bu türün daha fazla çalışılması gerektiğini ve literatüre kazandırılması gerektiğini düşünmekteyiz.

Cytotoxic Effect of Tricholoma fracticum Extract on Hep3B Cells

Abstract

Cancer constitutes one of the greatest threats to human health. Considering that drug resistance is one of the main problems in cancer treatment, studies on the effects of natural compounds on cancer cells are increasing rapidly. Edible mushrooms are considered as a functional and healthy food source because they are rich in nutritional values. Studies have shown that they have anticancer, anti-inflammatory, antioxidant, and antimicrobial effects. Tricholoma fracticum (Britzelm.) Kreisel, which grows naturally in many regions of our country and in the province of Elazığ, is an edible mushroom consumed as food. In this study, it was aimed to determine the cytotoxic effect of methanol extract of T. fracticum at different concentrations (1000µg/mL, 500 µg/mL, 250 µg/mL, 125 µg/mL and 62.5 µg/mL) against Hep3B cells at 24 and 48 hours. 3-(4,5dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) method was used to determine the cytotoxic effect. In the results obtained, the methanol extract of T. fracticum showed a significant cytotoxic effect at a concentration of 1000 µg/mL at the 24th (1.97±0.65%) and 48th hour (0.61±0.27%). The IC50 value of the methanol extract of the fungus against the Hep3B cell line was calculated as 166.76±7.65 µg/mL at 24 hours and 134.38±5.45 µg/mL at 48 hours. We think that the cytotoxic results of T. fracticum extract against Hep3B cell line may be important and this species, which is less studied among Tricholoma species, should be studied more and brought to the literatüre.
