IL18 -137 G/C ve -607 C/A Gen Polimorfizmleri ile Kronik Hepatit B Arasındaki İlişki

Abstract

Amaç: Hepatit B virus (HBV) enfeksiyonu ve enfeksiyonu takiben ortaya çıkan patolojik değişimler oldukça ciddi sonuçları olan global bir sağlık sorunudur. IL18, HBV gibi enfeksiyöz organizmalara karşı immun sistemin regülasyonunda oldukça önemli rol oynayan bir sitokindir. Yapılan son çalışmalar IL18 genindeki varyasyonların IL18 gen ekspresyonunu etkilediğini göstermiştir. Bu çalışmada IL18 gen polimorfizmleri (rs1946518, rs187238) ile kronik HBV enfeksiyonu arasındaki ilişki araştırılmıştır. 
Yöntem: Kronik hepatit B hastası olan 113 birey ve kontrol grubu olarak seçilen 71 bireyin IL18 gen polimorfizmi polimeraz zincir reaksiyonu-restriksiyon parça uzunluk polimorfizmi (PCR-RFLP) yöntemi kullanılarak belirlenmiştir. 
Bulgular: IL18 -137 G/C polimorfizmi için genotip frekansları hasta grubunda GG, GC ve CC genotipleri sırasıyla %33.6, %50.4 ve %15.9 kontrol grubunda ise %32.0, %46.5 ve %18.3 olarak saptanmıştır. IL18 -607 C/A polimorfizmi için ise CC, CA ve AA genotip frekansları hasta grubunda %55.8, %34.5 ve %9.7; kontrol grubunda ise %62, %31 ve %7 olarak bulunmuştur. IL18 polimorfizmlerinin genotip dağılımlarının gruplar arasında istatistiksel açıdan anlamlı bir farklılık göstermediği tespit edilmiştir (sırasıyla p=0.66, p=0.85).
Sonuç: Elde edilen bu sonuçlar IL18 gen -137 G/C and -607 C/A gene polimorfizmi ile kronik Hepatit B arasında anlamlı bir ilişki olmadığını göstermektedir. Ancak daha büyük ölçekli çalışmalar IL-18 -137 G/C and -607 C/A gene polimorfizminin kronik hepatit B patogenezinin anlaşılmasında faydalı olabilir.
