Cytotoxic and antioxidant effects of root extracts of Alchemilla mollis (Buser) Rothm

Abstract

Aim: It is known that the herba of the species belonging to the genus of Alchemilla L. is used as a diuretic, astringent, antispasmodic, curative, cure in invalid menstruation and for convulsive purposes. It has been determined that they have antioxidant, antimicrobial and anti-inflammatory effects due to their rich polyphenol content. This study, it was aimed to determine the effectiveness of the extracts prepared from the roots of the A. mollis, one of the species belonging to the genus, by performing various activity tests to determine the antioxidant and cytotoxic effects. Method: In this study, determination of total phenol and flavonoid amount of methanol extract prepared from roots of A. mollis species and sub-extracts with different polarity obtained by fractionation of this extract and, determination of radical scavenging effects of DPPH and ABTS to obtain their antioxidant capacity was evaluated. Cytotoxic effects of these extracts in L929 and MDA-MB-231 cell lines were also evaluated. Result: High phenolic content and accordingly high antioxidant capacity were determined in the root extracts of the plant. According to the results obtained from the extracts obtained from plant roots in L929 healthy mouse fibroblast cells and MDA-MB-231 breast cancer cell line; The extracts were found to be non-toxic to healthy cells within a certain concentration range, but toxic to breast cancer cells at certain doses. Conclusion: As a result, it has been determined with our studies that the A. mollis plant can be an important source in the discovery of promising therapeutic agents in cancer treatment by inhibiting the proliferation of cancerous cells with its rich phenolic compound content.

Alchemilla mollis (Buser) Rothm bitkisi köklerinden elde edilen ekstrelerin antioksidan ve sitotoksik etkileri

Abstract

Amaç: Alchemilla L. cinsine ait türlerin genellikle toprak üstü kısımlarının halk arasında diüretik, astrenjan, antispazmodik, yara iyileştirici, aşırı menstürasyonda kür olarak ve konvulsif hastalıkların tedavisinde kullanılmakta olduğu bilinmektedir. Sahip oldukları zengin polifenol içeriği sebebiyle de antioksidan, antimikrobiyal ve antienflamatuvar etkiler gösterdiği belirlenmiştir. Bu çalışmada ise cinse ait türlerden A. mollis bitkisinin köklerinden hazırlanan ekstrelerin antioksidan ve sitotoksik etkilerinin belirlenmesi için çeşitli aktivite testleri yapılarak, etkinliğinin tespit edilmesi amaçlanmıştır. Yöntem: Bu çalışmada A. mollis türünün köklerinden hazırlanan metanol ekstresi ve bu ekstrenin partisyonu ile elde edilen farklı polariteye sahip alt ekstrelerin toplam fenol ve flavonoit miktar tayini ve antioksidan kapasitelerini belirlemek için DPPH ve ABTS radikalini süpürücü etkileri tespit edilmiştir. Ayrıca ekstrelerin L929 ve MDA-MB-231 hücre hatlarında sitotoksik etkileri değerlendirilmiştir. Bulgular: Yüksek fenolik içeriğine bağlı olarak, bitkinin kök ekstrelerinde yüksek antioksidan kapasite tespit edilmiştir. Bitkinin köklerinden elde edilen ekstrelerin L929 sağlıklı fare fibroblast hücrelerinde ve MDA-MB-231 meme kanseri hücre hattında elde edilen sonuçlara göre; ekstreler belli bir konsantrasyon aralığında sağlıklı hücrelere toksik etki göstermezken meme kanseri hücreleri üzerinde belirli dozlarda toksik etkili bulunmuştur. Sonuç: Sonuç olarak A. mollis bitkisi zengin fenolik bileşik içeriği ile kanserli hücrelerin poliferasyonunu inhibe ederek kanser tedavisinde umut verici terapötik ajanların keşfedilmesinde önemli bir kaynak olabileceği tarafımızca yapılan çalışmalarla tespit edilmiştir

Our journal, which has been published via e-mail since its inception, has been published both online and in print. Following the Participation Agreement signed with TÜBİTAK-ULAKBİM Dergi Park in April 2015, it has started to accept and evaluate online publications.
