The present study was structured for molecular detection and characterization of Escherichia. coli (E. coli) isolated from cow’s raw milk samples. A total of 100 milk samples were collected in a way that 10 samples each were collected from markets of Jamalpur, Hasilpur, Khairpur, Qaimpur, Lal Sohanra, Lal Sohanra Park, Yazman Mandi, Rajkan, Ahmedpur East and Uch Sharif, district Bahawalpur, Pakistan. Milk samples were then cultured on different culture media for bacterial isolation. Identification of bacteria was done through Gram’s staining, properties of bacterial culture on different selective media and biochemical tests including catalase and coagulase tests. Final identification was performed through PCR and resolution of PCR products by gel electrophoresis. Out of 100 samples, 43 samples were found to be contaminated with E. coli. E. coli isolates were then amplified by 16S rRNA gene-based PCR. Antimicrobial sensitivity test was also performed to confirm the susceptibility of E. coli to different antibiotics. Results of the antimicrobial sensitivity test showed that the E. coli isolates were resistant to amoxicillin and erythromycin but sensitive to azithromycin, ciprofloxacin, gentamicin, norfloxacin and streptomycin.
