17α-Etinilestradiol ve 4-n-Nonilfenole Maruz Bırakılan İnci Kefali (Chalcalburnus tarichi, Pallas 1811) (Cyprinidae)’nin Karaciğer Hepatositlerinde Sitokrom P4501A’nın İmmunohistokimyasal Boyanmasındaki Değişimler

Öz
Bu çalışmada, 17α-Etinilestradiol (EE2; 1, 10 ve 100 ng/l) ve 4-n-Nonilfenolün (NP; 10, 60 ve 200 µg/l) nominal konsantrasyonlarına, semistatik koşullarda kronik olarak (32 gün), maruz bırakılan inci kefalinin karaciğer hepatositnde, sitokrom P4501A (CYP1A) enziminin hepatositlerdeki immunohistokimyasal lokalizasyonu, incelendi. Deneysel uygulamalar sonrasında, karaciğer kesitleri immunohistokimya yöntemi ile boyandı ve değerlendirildi. CYP1A immunreaktivitesi, kontrol grubuna ait bireylerde hepatositlerin sitoplazmasında, gözlenmedi. NP uygulanan gruplarda, CYP1A’nın immunohistokimyasal boyanmasında kontrol bireylerine kıyasla, bir farklılık belirlenmedi. EE2 uygulaması sonucunda, hepatositlerin sitoplazmasında nukleus memranından plazma membranına doğru artan ve orta şiddetli olan immunboyanma gözlendi. Elde edilen sonuçlar bu zenoöstrojenlerin, hepatositlerdeki CYP1A immunboyanması üzerine farklı etkiler gösterdiklerini ortaya koymuştur.
