Samsun İli Yaprak Lahana Üretim Alanlarında Görülen Virüslerin Belirlenmesi

Abstract

Yaprak lahana (Brassica oleracea var. acephala), ülkemizde Brassica cinsi içerisinde yer alan yaprağı yenen en önemli sebze türlerinden biridir. Ülkemizin en önemli lahana üretim alanlarından birisi konumunda olan ve üretimde ilk sırada yer alan Samsun ili Türkiye lahana üretiminin yaklaşık %30’unu karşılamaktadır. Lahanalarda virüs hastalıkları üretimi azaltabilmekte, kalite ve pazar değerlerini düşürebilmektedir. Samsun ilinde lahana üretim alanlarında virüslerin yaygınlığını belirlemek amacıyla 2013-2014 yıllarında sürveyler yapılmıştır. Arazi gözlemleri sırasında bazı yaprak lahana bitkilerinde mozaik, nekrotik lekeler, yaprak deformasyonları gibi karakteristik virüs belirtileri gözlenmiştir. Bu sürveyler sırasında, yaprak lahana üretim alanlarından 235 yaprak örneği toplanmış ve bu örnekler virüs-spesifik ticari poliklonal antiserumlar kullanılarak DAS-ELISA yöntemi ile Turnip mosaic virus (TuMV), Cauliflower mosaic virus (CaMV), Cucumber mosaic virus (CMV), Turnip yellow mosaic virus (TYMV) ve Beet western yellows virus (BWYV)’e karşı test edilmiştir. Serolojik testler sonucunda yaprak lahana örneklerinin %11.4’ünün CaMV, %7.6’sının TuMV ile enfekteli olduğu belirlenirken, %3.8 oranında CaMV+TuMV karışık enfeksiyonu tespit edilmiştir. Analiz edilen örneklerde CMV, TYMV ve BWYV ile enfekteli hiçbir örneğe rastlanmamıştır.

Determination of Viruses in Leaf Cabbage Production Areas in Samsun Province

Abstract

In Turkey, leaf cabbage which is an important leafy vegetable is one of the most economically important members of the genus Brassica. Samsun province is one of the greatest cabbage producing areas in Turkey and has nearly 30% of the cabbage production of Turkey. Virus diseases on cabbage reduce production and decrease quality and marketing value of the crop. Surveys were done to determine the distribution of viruses in cabbage cultivated areas in Samsun in 2013-2014. According to the results of field observations, characteristic virus symptoms, such as mosaic, necrotic spots, and leaf deformations were observed on some leaf cabbage plants. A total of 235 leaf samples was collected from cabbage fields in Samsun province and tested for the presence of TuMV, CaMV, CMV, TYMV, and BWYV by DAS-ELISA using virus-specific commercial polyclonal antiserum. The results of serological tests showed that 11.4% and 7.6% of these samples were infected with CaMV and TuMV, respectively. The mixed infection ratio including CaMV and TuMV in the samples was 3.8%. However, infections of CMV, TYMV, and BWYV were not determined in any sample.
