Antalya ili Demre ilçesi biber yetiştiriciliğinde Tomato spotted wilt virus (TSWV) ve Cucumber mosaic virus (CMV) etmenlerinin yaygınlığının belirlenmesi

Abstract
Amaç: Bu çalışmada Antalya ili biber üreticiliğinin yoğun olarak gerçekleştirildiği Kumluca ilçesinde serada yetiştirilen biber bitkilerinde TSWV ve CMV varlığının serolojik olarak belirlenmesi ve DAS-ELISA testi sonucunda pozitif olduğuna karar verilmesinde uygulanan kriterlerin doğruluğunun RT-PCR yöntemi ile karşılaştırılması amaçlanmıştır.
Materyal ve Yöntem: Antalya ili Demre ilçesinde örtü altı biber üretim alanlarında sörveyler yapılarak virüs benzeri belirti gösteren bitkilerden örnekler toplanmıştır. Örnekler CMV ve TSWV etmenlerine spesifik monoklonal ve/veya poliklonal antibody’ler kullanılarak DAS-ELISA testine tabi tutulmuştur. ELISA okuyucuda farklı absorbans değerlerine sahip bazı pozitif örneklere etmenlere spesifik primerler kullanılarak RT-PCR testleri uygulanmıştır.
Araştırma Bulguları: DAS-ELISA yöntemi sonucuna göre toplanan 82 örnekten 65 tanesinde (%79) en az bir virüs etmenine rastlanılmıştır. Sadece CMV olan örnek sayısı 10 adet (%12) olup sadece TSWV olan örnek sayısı 47 adet (%57) olarak tespit edilmiştir. 8 örnekte (%10) hem TSWV hem de CMV’nin karışık infeksiyon yaptığı belirlenmiştir. DAS-ELISA sonucunda pozitif olduğu tespit edilen örneklerin ELISA okuyucuda farklı absorbans değerlerine sahip pozitif sonuç veren örneklerde etmenlerin varlığı RT-PCR yöntemi ile doğrulanmıştır.
Sonuç: Biber örneklerinde TSWV etmeninin daha fazla bulunduğu CMV etmeninin ise fazla yaygınlık göstermediği belirlenmiştir.

Abstract
Objective: In this study, it was aimed to serological determination of the presence of TSWV and CMV on pepper plants grown in the greenhouses in Kumluca district where Antalya pepper production is carried out intensely and compare the accuracy of the criteria determining that positive applied in by DAS-ELISA with RT-PCR method.
Material and Methods: Samples were collected from virus-like symptoms showing plants by conducting surveys in greenhouse pepper production areas in the Demre district of Antalya province. Samples were subjected to DAS-ELISA test using monoclonal and /or polyclonal antibodies specific for CMV and TSWV. RT-PCR tests were applied to some positive samples which have different absorbance values in the ELISA reader by using specific primers.
Results: According to the results of the DAS-ELISA method, 65 of the 82 samples (79%) were found to have at least one virus. The number of samples infected only with CMV was 10 (12%) and the number of samples infected only with TSWV was 47 (57%). In 8 samples (10%) were found to have both TSWV and CMV mixed infections. The presence of the factors in the samples which have positive absorbance values in the ELISA reader were compared by RT-PCR method and the presence of the factors was confirmed.
Conclusion: It was determined that TSWV factor was found more in pepper samples and CMV factor did not show much prevalence.
